猪δ冠状病毒核衣壳蛋白两个新的B细胞表位的鉴定

  • 猪δ冠状病毒(PDCoV)是一种新发现的猪肠道冠状病毒,可引起猪尤其是新生仔猪腹泻和脱水。PDCoV核衣壳(N)蛋白是病毒颗粒中最丰富的蛋白质,在病毒生命周期的几个阶段中起着至关重要的作用。它可以使机体在PDCoV感染的早期产生高水平的抗体。然而,PDCoV N蛋白上抗原表位的特性仍然是未知的。本研究从重组PDCoV N蛋白免疫小鼠获得两种单克隆抗体6B7和7F8。免疫印迹试验和间接免疫荧光检测证实,两株单抗仅能识别PDCoV,与传染性胃肠炎病毒和猪流行性腹泻病毒无交叉反应。利用PDCoV N蛋白中一系列原核表达的肽段对单克隆抗体6B7和7F8进行了表位定位。PDCoV N蛋白的326-333氨基酸残基和251-276氨基酸残基分别是单克隆抗体6B7和7F8的最小抗原表位。3D结构模型显示Mab 6B7识别N蛋白的线性表位,而7F8可能识别N蛋白表面的构象表位。这些结果表明,PDCoV N蛋白的两个新表位可能具有抗原结构和功能特性,本研究生成的单克隆抗体可用于研究PDCoV N蛋白的功能和病毒感染机制。
