大蜡螟(Galleria mellonella)核型多角体病毒蛋白水解酶的研究

摘要
用广泛的 pH 和不同的离子强度、温度研究了大蜡螟核型多角体病毒的多角体降解动力学,证明在病毒中存在一种蛋白水解酶,最适 pH 是8.8。以硫酸铵沉淀和 Sephacryl 柱层析分离纯化,酶活力提高5倍,冷冻干燥得白色絮状的纯化酶。层析图谱上得到3个峰。峰1是酶活力主峰,峰2仅以产生可溶性蛋白、峰3仅以降解多角体为主。由这三个组分的共同作用,多角体降解,病毒粒子释放,感染发生。电泳证明,降解的多角体产生两条带,P36可能是多角体蛋白的主要结构多肽,酶的反应速度是酶浓度的涵数,绘图得一级动力学曲线。

Abstract
Dissolve kinetics for polyhedral matrix protein of Galleria mellonella nuclear polyhedrosis virus was investigated with wide range of pH,diff- erent ionic strength and temperature.A proteolytic enzyme was confir- med with the viral polyhedron.Its optimum pH for dissolved polyhedron is 8.8.
