实验性巨细胞病毒持续感染小鼠的干扰素诱生反应

  • C57BL/6小鼠经腹腔感染MCMV后,唾液腺可持续分离出病毒102天以上。在感染后6小时至32天内,血清中未能测到>1:4的干扰素活性。然而,MCMV感染小鼠的脾细胞在体外仍具有产生干扰素的能力。除MCMV感染后第5天对NDV诱生干扰素滴度略有低下外,其他感染组与对照组小鼠脾细胞对NDV诱生α/β干扰素与Con A诱生γ干扰素的滴度均无显著差异。感染鼠脾细胞与经紫外线灭活后的MCMV感染的同品系鼠胚细胞共培养也能产生干扰素,其高峰在MCMV感染后第9天。此种干扰素不耐pH2处理,经抗干扰素血清中和试验鉴定为γ干扰素。此外还进一步证明,尽管在感染后第5天的鼠脾平皿贴壁细胞(主要为巨噬细胞)中仍能分离到MCMV,但此细胞组分在体外用NDV诱生后产生的α/β干扰素滴度都与对照组无显著差异。因此认为:C57BL/6小鼠持续感染MCMV后,血清中未能测到干扰素,并非由于MCMV感染直接抑制鼠脾细胞产生干扰素反应的能力。整体动物在MCMV感染后干扰素反应低下可能与体内存在其他免疫调节性体液因子或细胞有关。
