荧光底物在酶联免疫吸附试验中的应用

  • 碱性磷酸酶底物,对硝基酚磷酸二钠(PNPP),在酶联免疫吸附试验(ELISA)中已普遍使用。本实验用一种荧光底物,4-甲基聚缴花酮磷酸酯做间接ELISA反应(FS-ELISA),同前者进行了比较。当用0.1ml的黄瓜花叶病毒(CMV),浓度为0.1ng/ml包被时,在紫外光下仍可看到反应所产生的荧光。FS-ELISA对CMV测定的敏感度比用PNPP底物时高100倍。最后对该方法用于定性和半定量检测抗原的问题进行了讨论。
