摘要
以NCM为固相载体、应用间接ELISA法测定了纯化的PVX、PVY和PVS;对接种的烟草,马铃薯块茎的芽、休眠块茎顶端的PVX、PVY和PVS也分别进行了测定。结果表明检测纯病毒PVX、PVY和PVS的灵敏度分别为2.76pg、2.835pg和5.831pg。检测感病烟草汁液马铃薯块茎芽、休眠块茎顶端的稀释度PVX分别为:1/20480~1/81920、1/20480和1/5120;PVY分别为1/81920、1/20480和1/5120;PVS分别为1/81920~1/327680、1/20480~1/81920和1/5120~1/20480。和Cocktail-ELLSA相比,检测PVX和PVY的灵敏度提高至少16倍。实验结果表明,Dot-ELISA是检测PVX、PVY和PVS灵敏而有效的方法。

Abstract
Purified potato virus Y, X, and S were detected by indirect enzyme-linked immunosorbent assay (ELISA) on nitrocellulose membranes. Potato virus Y, X, and S in infected tobacco, sprouts of potato tubers, rose end of dormant tubers were detected respectively. The results indicated that the small amount of sample required permit the detection of as little as 2.76pg, PVY, 2.835pg PVX and 5.83pg PVS in purified preparations.
