中国首例输入性裂谷热病例的病毒分离与系统进化研究

  • 裂谷热病毒主要分布在非洲东部和南部地区,埃及、沙特阿拉伯和也门也报告过感染病例。近年来,进出口动物贸易的显著增加提高了裂谷热病毒传播的风险。该论文使用实时荧光RT-PCR检测、病毒分离、全基因组测序和系统进化分析等方法,对中国首例自安哥拉输入的裂谷热感染病例进行了分析。实时荧光RT-PCR结果显示该病例血清样品裂谷热病毒核酸阳性。接种后的Vero和BHK-21细胞中观察到典型的细胞病变效应,且在细胞培养上清中检测到裂谷热病毒核酸。对分离的BJ01裂谷热病毒进行全基因组深度测序,并将全基因组序列提交到GenBank, L、M和S基因片段登录号分别为KX632066、KX632067和KX632068。Blast分析表明所有三个基因片段与2009年南非的Kakamas羊分离株序列密切相关。L片段的DUF3770功能域N277S和S307N发生氨基酸突变,RDRP结构域三个氨基酸位点有突变,M片段NSm蛋白的V17I和G1蛋白的I385M位点发生非同义突变。系统进化分析显示,该病毒株位于进化树E分支,是目前E分支上第一个报道的可感染人类的裂谷热病毒,推测可能由于BJ01病毒株某些重要和关键氨基酸位点突变导致了从动物到人的跨种传播。
