基于铁蛋白纳米颗粒展示Spike蛋白的ELISA方法的建立及其在猪丁型冠状病毒血清学监测与免疫评价中的应用

  • 猪丁型冠状病毒(PDCoV)是一种引起哺乳仔猪严重腹泻和死亡的新兴肠道病原体,给养猪业带来巨大经济损失。为建立可靠的血清学监测工具,本研究开发并优化了一种以铁蛋白纳米颗粒展示的刺突(S)蛋白为包被抗原的间接酶联免疫吸附试验(iELISA)。结果显示,该方法具有优异的诊断性能,批次间和批次内的变异系数均低于10%,且与常见猪病原体无交叉反应;与间接免疫荧光试验(IFA)的总体符合率达93.75%,且与中和抗体效价呈强正相关。临床应用检测发现,331份血清样本的PDCoV阳性率达48.64%,表明该病毒在检测地区广泛流行。综上所述,本研究建立的iELISA方法快速、敏感、特异且重现性好,适用于大规模血清流行病学调查和疫苗效力评价,为PDCoV的防控提供了重要技术支撑。
