小鼠腹水瘤病毒(SRSV)及其核糖核酸的纯化和某些性质的研究

摘要

从感染NIH3T3小鼠成纤维细胞上清液中分离小鼠腹水瘤病毒,用蔗糖密度梯度离心法纯化。从新鲜病毒中取小鼠腹水瘤病毒RNA,并用酚-氯仿抽提和蔗糖梯度分离。各组分的沉降系数(S)和分子量通过超速离心凝胶电泳分析测定。SRSV RNA主要由两组分组成,一是沉降系数约60～70S(热处理后约35S),还有少量18S和28S,另一是4～5S。35S组分在体外能指导无细胞蛋白的合成。

Abstract

SRSV was obtained from the supernatant of infected NIH 3T3 mouse fiblasts and purified by sucrose density gradient centrif ugation.SRSV RNA was prepared from fresh viral pellets and extracted with chloroform-phen(?)l and fractionated on sucrose gradients. The RNAs of various fractions were determined by sedimeptation coefficient(s) and followed by gel electrophoresis.
