Ib亚型赤羽病毒的分离鉴定及其在乳鼠上发病机制的研究

  • 迄今为止,中国分离的赤羽病病毒主要集中在Ia基因亚群和基因群II。在本研究中,我们用Vero细胞分离出了一株病毒,并通过PCR和间接免疫荧光试验确认为AKAV,命名为LK07株。LK07株与Ib基因亚群中的ISR-170/18、ISR-01、Aksu-1和Aksu-2株有较高的同源性。这是中国首次检测到赤羽病毒Ib基因亚群。为了建立哺乳小鼠的感染模型,我们将LK07病毒注射到小鼠的大脑、腹腔和皮下,并同时设立了正常对照组。脑内注射组的哺乳小鼠出现了停止吮乳、痉挛、抽搐和死亡等临床症状。组织切片结果显示脑组织局部结构疏松。qPCR和免疫组织化学结果显示所有感染组的哺乳小鼠脑组织均有阳性信号。这为进一步的AKAV分子流行病学和检测研究提供了一定的科学依据。
