一株可跨物种感染鸡的新型鹌鹑溃疡性肠炎相关腺病毒的鉴定与分析

  • 腺病毒感染对公共卫生和家禽养殖业构成重大威胁。唾酸酶腺病毒属(Siadenovirus)成员可感染鸟类、蛙类和龟类,多数毒株编码一种其他腺病毒中没有的唾液酸酶(sialidase)。火鸡出血性肠炎病毒(THEV)是一种唾酸酶腺病毒,可引起火鸡出血性肠炎与免疫抑制,使其易发生继发性细菌感染,给火鸡养殖业造成重大经济损失。本研究从表现为严重肠道出血、肾脏肿大发白等疑似溃疡性肠炎样症状的鹌鹑体内鉴定出一种新型腺病毒,并通过基因组比较与系统发育分析发现,该病毒归属于唾酸酶腺病毒属(Siadenovirus),与THEV毒株系统进化关系较近。该病毒编码的 DNA 聚合酶(Pol)氨基酸序列与所有已知腺病毒的同源性≤76.3%,其中与 THEV(NC_001958)的Pol同源性最高,为76.3%。这些结果表明,该病毒为一株新型唾酸酶腺病毒,命名为鹌鹑唾酸酶腺病毒(Quail siadenovirus),即鹌鹑腺病毒1型(QAdV-1),种学名暂定为 Siadenovirus coturnicis。进一步动物实验发现,QAdV-1 可在鹌鹑体内建立持续性感染并具有致病性,可见病毒广泛分布于宿主多种组织器官中,其中在肾脏、盲肠、法氏囊和脾脏中病毒滴度最高。另外,本研究发现QAdV-1可感染雏鸡,然而尽管在感染鸡的肛拭子和多个器官中均可检测到 QAdV-1,但感染鸡未表现出明显临床症状或死亡,且鸡的肛拭子和体内各组织中的病毒滴度均显著低于鹌鹑。鉴于QAdV-1 与THEV 基因组序列间的较高一致性(约74.5%),推测QAdV-1也可能诱导鹌鹑免疫抑制,进而增加其继发大肠杆菌病/梭菌病的易感性。总之,本研究发现QAdV 1 是一种溃疡性肠炎相关病原体,存在跨物种传播风险,后续有待进一步对其致病机制、潜在的免疫抑制通路及针对性疫苗的研发进行深入研究。
