单纯疱疹病毒空斑形成影响因素的研究

摘要

使用pH 7.0的2％小牛血清Hanks氏液稀释Ⅰ型单纯疱疹病毒(冰盒存放6天内),接种原代人胚肌皮单层细胞瓶0.5ml,37℃静止吸附120分钟,弃去液体,不经洗涤或换塞,再加入2％小牛血清、0.5％水解乳蛋白与1个单位抗血清的维持液,培养46天后,分别取1％石碳酸复红与龙胆紫-甲醇进行二次染色,形成的空斑用肉眼清晰可见。同型抗血清可抑制空斑产生,表明本法具有特异性。本工作已用于抗病毒药物的筛选。

Abstract

In the present study we have first identified the factors regulating the formation of plaques by herpes simplex virus (type 1) infecting the primary muscle and skin monolayer cells of human embryo. The numbers of plaques formed in the two varieties of viral diluted solutions(e.g., Hanks solution containing 2％ fetal bovine serum and/or 199 solution)are parellel to each other.
