摘要
高灵敏度的蛋白质银染色法最初是由Switzer和Merril于1979年建立的,其后Oakley、入江伸吉等人加以改良和简化,使检测的灵敏度提高到ng水平,可与放射自显影媲美。后来Somerville和Wang将该法扩大应用于聚丙烯酰胺凝胶中DNA和RNA的染色,山崎健一等又将该法推广到琼脂糖凝胶中DNA和RNA的染色。银染色法的突出优点是灵敏度高,用于蛋白质染色,比考马斯亮蓝染色感敏约50—100倍;用于RNA染色,比亚甲蓝染色敏感100—1000倍;用于DNA染色,比溴化乙锭染色敏感10—20倍。现将该法的要点和笔者的经验介绍如下。
