SHANG Jia-Ming, YUAN Yun-Hua, CHENG Hong

1992, 7(2): 124

Up to present, few was reported about the natural infection of murineeytomegalovirus(MCMV) in laboratory mice. Based on the experimental infection of MCMV, we made a survey on the natural infection of MCMV in 10 common used strains of mice It was found that of 334 samples of submaxllary gland homogenate, none were positive for viral isolation, whereas specific anti-MCMV antibodies were found in 10.9-87.50% of different strains of mice. There were tendencies of increase in antibody positive rates as the mice...

1992, 7(2): 134

Hepatitis E virus(HEV) was isolated in human embryomic lung cell inoculated with liver tissue suspension from an HEV-infected rhesus monkey, Slight cytopathic effect(CPE) appeared at the 27th day after 4 blind passages of viruses in cell culturs to at 17th day after 7 passages. Low amounts of 27—34nm virus-like particles(VLPs) in cell cultures were visualized by immunoelectronmicroseopy(IEM). 2 of 3 rhesus monkeys inoculated with 20ml of pool of cell cultures from 4 to 7 passages developed significant eleva...

JIANG Ke-Qiang, PENG Qin-Lin, E Hai-Chun

1992, 7(2): 138

The method of using ABC-ELISA to measure anti-HBC in the serum of patients with hepatitis B was developed. The experimental results indicated that the sensitivity of ABC-ELISA was five times higher than that of the conventional ELISA. On the average, the positive detection rate of anti-HBc by means of ABC-ELISA increased by 42.42% over the one by using conventional ELISA. Repeated experiments showed good reproductivity. Alternative tests using various antigens and antibodies of HBV and blockage test using p...

Half and one year after the epidemic of acute hemorrhagic conjunctivitis(AHC) caused by eoxsackievirus A24 variant(CA24v) in the autumn of 1988, 423 sera were collected and the neutralization antibody(NA) and specific IgM against CA24v were tested in microplate with Hela cells. The positive rate of NA was 37.9% in the spring and 48.8% in the autumn of 1989. The specific IgM was 3.3% in the spring and 6.7% in the autumn of 1989 and one strain of CA24v was isolated from a cold-like patient at this time. The r...

YAN Jia-Xin, HUANG He, BO Nan-Sheng, YAO Jie

1992, 7(2): 149

A sensitive and simplified method of ~(125)I-labeled peptide mapping was used to estimate the rates of the haemagglutinin(HA) mutation of 5 strains of influenza Al viruses and 5 strains of influenza A3 viruses. It could be calculated that the average rates of HA mutation for these two subtypes of viruses are 0.47% and 0.44% per year respectively. The strains isolated in 1986 showed some specialities as compared with the average rates of HA mutation The relation between the rates of HA mutation and the epide...

YU Yong-Xin, YAO Zhi-Hui, AN Qi

1992, 7(2): 156

Serotypic classification of 14 HFRS virus isolates from different sources were studied using plaque reduction tests(PRNT) sera of rabbits inoculated with different HFRS viruses were tested against reference type-1 Hantaan virus and type-2 Seoul virus. The results indicated that except one strain(K_(24)), all the 13 isolates could be classified into either serotype 1 or serotype 2 according to the differences(≥4 fold) of the neutralizing titers between the sera reacted against reference type 1 and type 2 vir...

NIU Jian-Zhang, GUO Ge-Jian, Edwards P., Bowden D.S.

1992, 7(2): 160

A RNA hyhridization assay for hepatitis D virus in serum and liver specimens, from human and infected chimpanzee experimentally, was described using a nonradioactive Digoxigenin labelled RNA probe, and was compared with the ~(32)P labelled RNA probe. Findings show that the sensitivity and specificity of the two probes are the same(about 0.2pg of homologous DNA). The results indicated that Digoxigenin labelled RNA probe can be used for efficacious detection of HDV RNA in serum and liver tissue from human or ...

CHEN Sheng-Min, GUO Hui-Yu

1992, 7(2): 166

In this study we developed a method for characterization of the leetin binding properties of D2V by a panal of lectins with different sugar specificity. The dengue-2 virus(D2V) (New Guinea C strain) was cultured in Aedes alhopietus C6/36 cells. The virus-speeific and cellular proteins of the infected and mock-infected cell lysates were separated by diseontinous SDS-PAGE and transferred electrophoretically onto NC sheet. The blots were then probed with 12 kinds of biotinylated leetius(Con A, PSA, LCA, SBA, D...

1992, 7(2): 176

In this study, expanded human trial with M, Gerbil kidney tissue culture inactivated HFRS vaccine was carried out and neutralizing antibody response was assessed by plaque reduction neutralization(PRNT) and CPE neutralization(CPENT) methods. According to the data of all 74 person immune sera assayed by the two methods, the rates of seroconversion and GMT tesed by CPENT were significantly higher than that by PRNT. Several vaccinating groups were studied and the neutralizing antibody levels were as follows: v...

The BALB/C mice were immunized with HeLa cells infected with respiratory syncytial virus(RSV, Wuhan strain). The immunized spleen cells were fused with SP2/0 myeloma cells which were obtained from the SP2/0 solid tumor grown in BALB/C mice. Thirteen hybridoma cell lines secreting antiRSV monoclonal antibody(MeAb) were established. The chromosome number of these cell lines was ranged from 94—104 and the seerated MeAbs belonged to routine IgG1, IgG2a, IgG2b subclasses. The ascites antibody titer was from 1: 1...

Recently, we isolated a non-occluded virus from spontaneous mortal Periplaneta fuliginosa(Serville). The virus suspension was mixed in sterile food to feed healthy P. fuliginosa nymphs(of 154—169 days). The percent mortality of the nymphs which died with infected virus was over 98%. Under electronmicroscope the virus was isometric particle about 23nm in diameter. The ultraviolet absorption of the virus was as character as nucleoprotein Purified virus suspension was used to distinguish the nucleic acid kinds...

ZHANG Qin-Feng, ZHAO Yu-Xia, YANG Ying, JIN Xin-Zao, SHU Xiang-San

1992, 7(2): 192

It had been manifested that corn was readily infected by wheat BYDV The symptoms were similar to those of the wheat The most effective veetor aphid species was R. padi, while M. avena, S. graminnm and R. maidis were also among the vectors. The isolates belongs to the main strain of BYDV, eg. S. graminum/R. padi(GPV) strain. Mutual transmission in the fields between wheat and corn was evident.

LU Guan-Cheng, HONG Jian, XU Zheng, HU Shi-Meng

1992, 7(2): 197

In Zhejiang, Jiangsu and other provinces, 5 varieties of the garlic crops are totally diseased. The virus disease frequently causes degeneration of viruliferousness, loss of yields and reduction of tolerant to store. Symptoms due to the infection of virus are chlorotic mottling between the veins, yellow streaking and mosaic, scarcely visible and leaf distortion or deformation. The virus can be artificially inoculated on Vicia faba, then produces brown local lesions, slight spot on Chenopodium murale, necrot...

Three strains of retrovius, which can give positive results both by ELISA and immunofluoreseenee assay(IFA) with SRV-1 newly have been isolated from Rhesus monkeys(Macaca mulatta) catehed frim three areas of China. It have been confirmed by Western Blot that their seroantibodies response with SRV-1 positively. One of them occurs cross-response with of SRV-1 and STLV. Lots of the virus particles having typical ultrastuctures of retrovirus have been observed by electron microscope.
