A rapid method for detection of HBV-DNA was developed. This assay was based on the hybridization of biotinylated oligonucleotide probe to target DNA immobolized on nitrocellulose membrane which was then incubated with avidin-alkaline phosphatase conjugator and measured with chromogenic substrates. The practicality of the method was investigated. It was found that the method was fast (4hr),sensitive(1-10pg)and expedient for laboratory and clinical detection of HBV-DNA.

A total of 7.2 kb of full-length genome of hepatitis E virus (HEV)XT-179 strain was cloned as cDNAs that isolated from a HE patient s fecal sample during an epidemic outbreak peak of Sept. 1990 in the East Xinjiang of China, and the complete nucleotide sequence has been determined. Antisense phosphorothioate oligodeoxynueleotides respectively complementary to initial region of 2.1kb mRNA(oligomer A,14nts)and enhancer I(oligomer B,12nts)of HBV were synthesed by chemi cal synthetic method,and inhibition of these oligomers on expression of HBsAg and HBeAg in cell cul ture system was studied. The levels of interleukin-1 and interleukin-2 production in 16 healthy blood donors bone mar row mononuclear cells infected by HBV in vitro and on 16 cases with chronic hepatitis B were simulta neously assayed. In this assay,we studied the possibility of raccoon pox virus (RCN) TK gene as a viral selection marker on Rat-2 cells. Southern blot showes that RCN has TK gene sequence though it hybridizes week ly with vaccinia virus TK gene. But RCN is resistant to 5-BUdR even after it was plaque purified in HAT media.

Properties of Polyhedron Protein from Leucaia Separata Nuclear Polyhedrosis Virus and Its Inhibition to Growth of Several Tumor Cells

SDS-PAG electrophoresis showed that the polyhedron proteins of Leucaia separata nuclear polyhedrosis virus(LsNPV)was consisted of several polypeptides, the major band of MW 32kD was the major structural polypeptides of polyhedra. Amino acid composition of 32kD protein was analyzed after purification by chramatography of Sephacryl S-200 column. The cell growth of three tumor (Hela, HLAMP, HICAM)was inhibited by 32kD protein, DNA replication labeling with ~3H-TdR recognized the result.

The temperature-inducible expression vector containing the beet necrotic yellow vein virus(BNYVV)isolate NM coat protein gene was constructed and transferred into E. coli DH5 . High-level expression of the specific protein was achieved by temperature induction. The results of SDSPAGE and Western blotting show that the expression product which accumulates 19.5% of the total cellular proteins estimated by Shimadzu CS-910 scanning is 21kd BNYVV coat protein.

Tai MV was routinely detected and diagnosed by ELISA method. The detective sensitivity of the method was 4ng. Fields detective results showed that the soil, the roots tubers of Taizishen, symptomless plants and the surfaces of seeds had serological reactions with Tai MV antiserum. And it was confirmed that Taizishen seeds could eliminate Tai MV completely by infusing in 10% Na_3PO_4 solution for 20 minutes.
