The vertical transmission of epidemic haemorrhagic fever virus (EHFV) in mice has not been confirmed. From 1985 to 1986, We conducted the studies in epidemic fiels for getting a reliable answer to the above problem on the basis of a serial sludies in the laboratory. In our study IFA was adopted to detect the EHFV antibody and antigen; tissue culture technique was adopted to isolate EHFV from fetuses which were dissected from the uteri of infected mice. Our results are as follows : In the field study, the sp...

JIN Yu-Huai, WANG Yu-Kun, GU Bao-Liang

1989, 4(4): 345

Rubia cordifolia is a traditional Chinese drug. In this paper, it was reported that the methanol extract of this drug showed significant antiviral effect against herpes simplex virus 2 in vitro. In Vero cell line, virus inhibiting index reached 3 log. Virus reproduction inhibiting test showed that the reproduction of infectious virus parlicles was inhibited. By comparing the antiviral effect of this agent in different cell line, it was found that the antiviral effect of this extract was cell line dependent.

CHEN Huo-Sheng, GUO Hui-Yu, LUO Jin-Xian, HU Jin-Xin

1989, 4(4): 365

Recently we have prepared large amount of purified pEH 920 DNA which is inserted by complementary DNA of dengue virus 2 RNA. Using the [a-~(32)p] dCTP labelled pEH 920 DNA as the probe, dengue virus RNA in the supernatants of infected C6/36 mosquito cells have been detected by DNA-RNA dot-spot hybridization. The results show that the probe strongly reacted with dengue 2 virus RNA though it also cross-hybridized with dengue 1, 3 and 4 virus RNA. The preliminary experiments show that the probe can at least de...

LI Wei-Qi, WU Er-Ni-Sha, TUN Zu-Yin, CHEN Lu

1989, 4(4): 390

The assembly processes of the Leucoma selicis NPV are observed by electron microscopy of sections of infected larval fat body cells. The synthesis and envelopment of the virus nucleocapsid and stages in the formation of the polyhedral inclusion bodies are described. Bundles of fibrous material were seen in the nuclei and cytoplasm of cells contaning polyhedral bodies. We also found that in the same polyhedron there are single and multiply embedded virus rods.

1989, 4(4): 408

A virus isolate (w) has been obtained from the Oker river in W.Germany. The isolate was identified as Carnation ringspot virus (CarRSV) by means of inoculation on the test plant, electron microscopy, dot blot hybridization and serology. The molecular weight of the coat protein is 3.8×10~4. Northern blot analysis revealed that the two RNA species of the virus which consisted of c. 3.7 and 1.5kb, respectively, but without base sequence homology.

Effects of Temperature on ToMV Multiplication and Changes of Soluble Leaf Proteins in Tomato

XU Ren-Lin, YI Qiong-Hua

1989, 4(4): 415

ToMV concentrations in virus inoculated tomato plants were analyzed by local lesion test. The results showed that virus concentrations in resistan-t line GCR-267 was 1/40—1/50 that in susceptible line GCR-26. ToMV multiplication was strongly inhibited by gene Tm-2~a, which was not influenced by temperature. ToMV multiplication in GCR-26 was dependent on temperature, the higher the temperatature, the less the virus concentration. The changes of the soluble leaf proteins of tomato plants grown at constant or ...

1989, 4(4): 375

Isolation and characterization of a H isolate of enterovirus type 71 (E71) from the vesicle fluid of hand-foot-mouth disease have been reported in present study. The H isolate can grow at primary human embryo lung cells, MA104 cells, and BSC cells and results in a typical enterovirus cytopathic effect. 1-day-old mice inoculated ip with H isolate showed limb paralysis and died at 6th day postinfection. Numerous mature virus particles with a diameter of 25 nm in crystal arrays in the cytoplasm have been visualized under microscope. The paired sera of the patient have been tested and showed a 4-fold increase of neutralizing antibody to H isolate. Using 100 antibody units of anti-Cox A5, 7, 9, 16, E70, E71, and 50 antibody units of enterovirus LBM typing pools A-H as well as McAb p27 of E71 BrCr strain, H isolate could be only neutralized by both anti-E71 polyclone and monoclone antibodies. The minimal effective antibody units to H isolate are required respectively 1.6 for anti-E71 polyclone antibody and 64 for McAb p27. Furthermore, high titers of anti-Cox A16 have also showed some cross reaction to H isolate shares a common antigen with Cox A16.
